Description
This product is supplied as a concentrated solution containing Nitro blue tetrazolium (NBT) and 5-Bromo-4-chloro-3-indolyl phosphate (X-Phosphate p-Toluidine Salt) for Western Blotting. When this product is reacted with alkaline phosphatase (ALP), a blue-purple precipitate appears.
Direction for Use
1: Incubate a blotting membrane with an ALP-conjugated antibody and wash the membrane.
2: Dilute NBT/ X-Phosphate p-Toluidine Salt solution (Product No. N1113) to 1X before use.
(Dilute solution : 20 mM Tris-HCl pH 9.5, 2mM MgCl2 and 100 mM NaCl)
3: Incubate the washed membrane with 1X NBT/ X-Phosphate p-Toluidine Salt solution until color development.
4: Add deionized water to stop the color development.
Storage
The product should be stored at 4 ºC. Don’t freeze because of arising precipitate.
