Cyclothiazide: A Benzothiadiazide Antagonist of AMPA Receptors
α-Amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (also known as AMPA receptors) are a class of ionotropic glutamate receptors, and major players in mediating basal synaptic transmission and synaptic plasticity in the mammalian CNS. AMPA-receptor modulation could be a way of adjusting the excitatory–inhibitory balance. Therefore, AMPA receptors are emerging as a new target for epilepsy, depression, etc. Cyclothiazide has been reported as a benzothiadiazide antagonist of AMPA receptors, and a promising tool for analysis of allosteric regulatory mechanisms at AMPA receptors.
