Red 2X Sample Buffer for Differentiating Protein Samples by Color
In SDS-PAGE, the sample buffer containing Tris-HCl buffer, glycerol and sodium dodecyl sulfate (SDS) is utilized to optimize the charge of the protein sample.1) 2X SDS-PAGE sample buffer phenol red (1) is a red 2X sample buffer in which phenol red is added as a dye to confirm the progress of electrophoresis.2) The combination of both 2X sample buffer with bromophenol blue as a dye and 1 helps to prevent mistaking the samples by color (Figure). In addition, 1 has the advantage of making it easier to see the applied lane without overlapping the samples of the same color by changing colors of the sample buffers on the front side and opposite side when setting two gel plates in the electrophoresis tank.
The sample and 1 are mixed 1:1 when preparing the sample for electrophoresis. Since 1 does not contain any reducing reagents, a reducing reagent such as 2-mercaptoethanol or DL-dithiothreitol is added if necessary just before use.
