Application
Ifosfamide: An Oxazophosphorine Antitumor Alkylating Agent
Ifosfamide is an oxazaphosphorine alkylating agent with a broad spectrum of antineoplastic activity. It is a prodrug that requires metabolic activation by hepatic cytochrome P450 isoenzymes to exert its cytotoxic activity. The exact mechanism of action of ifosfamide has not been determined, but among various metabolites, isophosphoramide mustard probably represents the most important cytotoxic compound able to produce irreparable cross-links between DNA strands caused by alkylation by the isophosphoramide mustard at guanine N-7 positions. In addition, mesna [M0913] is often used as a prophylactic agent for side effects of oxazophosphorine antitumor alkylating agents such as cyclophosphamide [C2236] and ifosfamide.
