Application
JZL 184: A Potent and Selective Inhibitor of Monoacylglycerol Lipase (MAGL)
Endogenous cannabinoids, such as anandamide [A3448] and 2-arachidonoyl glycerol (2-AG) [A3490], are biologically active lipids that activate the cannabinoid receptors CB1 and CB2. Cannabinoid receptors, located throughout the body, are part of the endocannabinoid system (ECS) which is involved in a variety of physiological processes. Endocannabinoid signaling is terminated by enzymatic hydrolysis, a process that for anandamide is mediated by fatty acid amide hydrolase (FAAH), and for 2-AG is thought to involve monoacylglycerol lipase (MAGL). JZL 184 is a potent and selective inhibitor of MAGL and has been reported to raise mice brain 2-AG levels by eight-fold without altering anandamide and to attenuate neuropathic pain.
