Application
Teriflunomide: An Immunomodulatory Compound
Teriflunomide (A77 1726), the active metabolite of leflunomide [L0250], is an immunomodulatory compound. Teriflunomide has two activites: inhibition of pyrimidine nucleotide synthesis and interference with tyrosine phosphorylation. More specifically, it inhibits the enzymic activity of dihydroorotate dehydrogenase (DHO-DHase) which catalyzes the fourth step in the biosynthesis of pyrimidine, and tyrosine kinases which transfer a phosphate group from ATP to a protein in a cell. Teriflunomide reduces the proliferation of T and B immune cells that are active in multiple sclerosis (MS). For your reference, [C3217] is a synthetic intermediate.
