Application
Hydrazide Useful for Construction of Heterocycles
Experimental procedure: 1,3-Diphenyl thiourea (0.5 g, 2.19 mmol) is added to a stirred solution of IBX (0.65 g, 2.19 mmol) and triethylamine (1 mL, 7.11 mmol) in DMF (10 mL) for 5 min. Then, formohydrazide (0.45 g, 7.04 mmol) is slowly added to the reaction mixture with continued stirring for 3 h at rt. After completion, the reaction mixture is poured into a saturated solution of NaHCO3 (20 mL). The aqueous layer is extracted with ethyl acetate (3 × 15 mL). The combined organic layers are dried over Na2SO4 and evaporated to afford the crude product, which is purified by column chromatography on silica gel (petroleum ether : ethyl acetate) to afford 0.37 g of 1 as a white solid in 72% yield.
