Description
This kit consists of three components; 5,5'-Dithiobis(2-nitrobenzoic Acid) (DTNB, Ellman’s reagent), N-Acetyl-L-cysteine (NAC), and Diluent Buffer. Reaction of DTNB with SH groups causes the solution containing them to turn yellow in color. Use of an N-Acetyl-L-cysteine standard curve allows for the quantification of SH group concentration via absorbance measurement at 412 nm.

Store at 4 °C. Use within 6 months.
Direction for Use
- Prepare an SH group-containing sample, using Diluent Buffer to increase the volume to 200 µL if necessary.
- Add 1 mL of Diluent Buffer to 1 vial of N-Acetyl-L-cysteine to prepare a 10 mM NAC standard solution. Dissolve carefully by vortexing.
- Preparation of an NAC dilution series:
Dilute the 10 mM NAC standard solution prepared in step 2 10-fold with Diluent Buffer to make a 1000 µM NAC solution (of greater than 400 µL); further dilute this solution two-fold six additional times using Diluent Buffer such that you obtain seven samples (each greater than 200 µL in volume). The dilution series comprises these seven samples, along with a blank containing only Diluent Buffer. The concentration range this dilution series offers is appropriate in many cases, however if you plan on measuring a more dilute sample, adjust concentrations as needed. - Aliquot 100 µL of each sample or NAC solution to each of two wells of a 96-well plate.
- Prepare DTNB solution by adding 1 mL of Diluent Buffer to 1 vial of DTNB. Dissolve carefully by vortexing. (Use DTNB solution as quickly as possible following preparation.)
- Add 15 µL of this DTNB solution to each well.
- Incubate at room temperature for 10 minutes.
- Measure the absorbance at 412 nm. (Broad range measurement from 380 - 440 nm is also possible.)
- Create a standard curve by plotting the absorbance of each standard solution against its SH concentration in μM. Use this standard curve to determine the SH concentration of your samples.
