10 µL of 25 mM labeling reagent 1 in dry benzene is mixed with 10 µL of 0.5 mM androsterone in dry benzene (containing 0.5 mM quinuclidine*), and incubated at 60 °C for 30 min. The reaction solution is quenched with 980 µL of 50% acetonitrile solution containing 1% acetic acid, use it as the HPLC sample solution.
*For primary alcohols, quinuclidine is not necessarily needed.
