The Decoloring Solution for Plant Tissue-Clearing Method iTOMEI

Tissue-Clearing Reagent iTOMEI-D [for Plants] (1) is a decoloring solution used for the improved TOMEI method (iTOMEI), a transparency method that is an improved version of the TOMEI method developed by Matsunaga et al.^1,2) iTOMEI has the advantage of making plants transparent while suppressing the quenching of fluorescent proteins such as GFP and tdTomato. Plants that have been fixed in 1% paraformaldehyde / PBS can be immersed in the decoloring solution 1 to remove chlorophyll from plants. The transparent plants can then be observed by treating them with a mounting solution that matches the refractive index of the objective lens to make the refractive index uniform.
