初のフルオロキノロン系合成抗菌剤,ノルフロキサシン
Norfloxacin is the first fluoroquinolone antibacterial reagent which is synthesized from [C2897] with piperazine. Its structure is related to nalidixic acid [N0490] but its potency has been increased by a fluorine atom and by a piperazine moeity. Thereby, norfloxacin has a broad spectrum of activity against both Gram-positive bacteria and Gram-negative bacteria. Norfloxacin and the other fluoroquinolones inhibit bacterial DNA synthesis by inhibiting the enzyme gyrase which catalyzes the operations of the DNA double strand such as opening, under winding, and closing. Fluoroquinolones appear to prevent specifically the resealing of opened DNA strands and thereby act bactericidally.
