Gatifloxacin: An 8-Methoxy Fluoroquinolone Antibacterial Agent with a Broad Antibacterial Spectrum
Gatifloxacin (AM-1155) is an 8-methoxy fluoroquinolone antibacterial agent with a 3-methylpiperazinyl substituent at C7. Fluoroquinolone fungicidal antibacterial agents are inhibitors of bacterial DNA gyrase and topoisomerase IV.1,2) DNA gyrase is an essential enzyme involved in the replication, transcription and repair of bacterial DNA. Topoisomerase IV is an enzyme known to play a key role in the division of chromosomal DNA during bacterial cell division. As just described, the mechanism of action of fluoroquinolones is different from that of other antibiotics. Therefore, there is no cross-resistance between fluoroquinolones and the other classes of antibiotics. In particular, gatifloxacin has a broader spectrum of antibacterial activity than the older fluoroquinolones and shows good activity against many Gram-positive and Gram-negative bacteria, atypical organisms and some anaerobes.3) In addition, gatifloxacin has higher selectivity for bacterial type II topoisomerases.4) (The product is for research purpose only.)
