Gefitinib: A Potent and Selective Inhibitor of EGFR Tyrosine Kinase
The epidermal growth factor receptor (EGFR), a receptor tyrosine kinase (RTK), is expressed on the cell surface of both normal and cancer cells and plays a role in the processes of cell growth and proliferation. It is reported that gefitinib (ZD1839) is a potent and selective inhibitor of EGFR tyrosine kinase (EGFRTK). In vitro, gefitinib arrests the cell cycle at G1 phase by inducing intrinsic cyclin-dependent kinase (cdk) inhibitors. Furthermore, gefitinib inhibits vascular endothelial growth factor (VEGF) production in tumor cells through inhibition of EGFR signaling, leading to suppression of angiogenesis.
