A769662 (1) is a cell-permeable activator of AMP-activated protein kinase (AMPK).1) 1 was found to directly stimulate partially purified rat liver AMPK with EC50 = 0.8 μM and inhibited fatty acid synthesis in primary rat hepatocytes with IC50 = 3.2 μM.1)
1 provided significant stimulation to purified rat liver AMPK (EC50 = 116 nM) and stimulation of the kinase by 1 was 50 times more potent than that by AMP.2) The mechanism of activation of AMPK by 1 is allosteric and involves inhibiting dephosphorylation of the kinase on Thr-172.3) 1 inhibits reprogramming of somatic cells into induced pluripotent stem cells (iPSCs)4) and proliferation of mesenchymal stem cells.5)
