Application
Doramapimod: A Highly Potent Inhibitor of p38 MAPK
The p38 mitogen-activated protein kinase (p38 MAPK) is a Ser/Thr kinase, originally isolated from lipopolysaccharide-stimulated monocytes. It plays a crucial role in regulating the production of pro-inflammatory cytokines, such as tumor necrosis factor (TNF) and interleukin-1.1) Therefore, blocking this kinase may offer an effective therapy for the treatment of many inflammatory diseases. Doramapimod (BIRB-796) is a highly potent inhibitor of p38 MAPK that blocks TNFα release in LPS-stimulated THP-1 cells.2) Furthermore, doramapimod is also used in regenerative medicine area. Doramapimod enhances the stem cell activity of cultured umbilical cord blood-derived hematopoietic cells cultured in serum-free medium supplemented with SCF, TPO, and FLT3L.3) Doramapimod also blocks the osteogenic differentiation effect of myocilin on human mesenchymal stem cells (HMSCs).4) (The product is for research purpose only.)
