Description

  • Biofilms are highly organized structures of microbial communities composed of a matrix of polysaccharides and other substances secreted by microbes and/or aggregates of microbial cells. Microbial cells become resistant to antimicrobial drugs and host defense systems when they form biofilms. Clearing Reagent iCBiofilm-D has been developed to allow for live-cell-imaging of biofilm at thicknesses of more than several tens of micrometers. Simple addition of iCBiofilm-D solution to a biofilm renders it transparent within several seconds, allowing for easy observation using light microscopy techniques. This product has been filter-sterilized.

Note

    • For Research Use Only. Not for Diagnostic or Therapeutic Use.
    • Vigorous manipulation of this product may cause biofilm to peel off.
    • 24-hour live cell imaging of biofilm can be observed.
    • If clear images cannot be obtained or biofilm formation is prevented in iCBiofilm-D, try diluting iCBiofilm-D twice by culture medium.

Directions for Use (for live-cell-imaging of biofilm)

    1. Prepare a live-cell-imaging medium. Dissolve the culture medium powder in iCBiofilm-D to create a biofilm culture medium by autoclaving and so on.
    2. Prepare a biofilm in a glass bottomed culture dish (35mm diam.) in ordinary medium (not live-cell-imaging medium). Add staining reagents as needed. Culture conditions (including medium, temperature, time, etc.) all play roles in biofilm formation; the conditions specific to your microbe of interest must be determined separately.
    3. After removing the culture medium, wash the dish with the ordinary medium to remove planktonic cells.
    4. After wash, add the live-cell-imaging medium. Add staining reagents as needed.
    5. Observe the biofilm and obtain the images of live-cell-imaging.
