Application
Methotrexate: An Inhibitor of Dihydrofolate Reductase (DHFR)
Methotrexate (MTX), analog of folic acid, is an inhibitor of the enzyme dihydrofolate reductase (DHFR) which catalyzes the conversion of dihydrofolate to the active tetrahydrofolate, leading to interfere with folic acid metabolism. Result is inhibition of DNA synthesis and cell proliferation in S phases (the phase of DNA synthesis). Also, MTX has immunosuppressive and anti-inflammatory activity. Although no single mechanism is sufficient to account for all the anti-inflammatory activities of methotrexate, the release of adenosine from cells has been demonstrated. MTX is commonly used for the treatment of certain cancers including leukemia, rheumatoid arthritis, Crohn’s disease and psoriasis. (The product is for research purpose only.)
