Description

• 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyltetrazolium Bromide (MTT) is a tetrazolium salt that is converted to formazan when taken up by living cells. Although formazan forms crystals in cells due to its insolubility in water, it is soluble in DMSO. By dissolving these formazan crystals in DMSO and reading the resultant absorbance at 540 nm, an estimate of cell number can be obtained. This product comprises a solution of 5.0 mg/mL MTT dissolved in PBS. To use, simply apply 1/10 of the culture medium volume of MTT Solution to live cells.

Storage

• Stored at -20 °C and protected from light. Avoid repeated freezing and thawing.

Direction for Use

• MTT solution can be added at any point in the cell culture. For best results, it should be added to cells in the logarithmic growth phase. Pre-determine the number of cells and seed 5,000 to 50,000 cells per well in a 96-well plate.

• 1. Seed 100 µL of cells in a 96-well plate.
  2. Incubate overnight in an incubator.
  3. Add 10 µL of MTT solution to each well.
  4. Incubate for 2-4 hours for color reaction.
  5. Remove the medium, taking care not to suck up the formazan.
  6. Add 100 µL of DMSO (Product No. D0798) and dissolve the formazan.
  7. Measure the absorbance at 540 nm.
