Description
Acridine orange is a cell membrane-permeant nucleic acid cell stain, which is incorporated into double-stranded DNA at a ratio of one to three base pairs. It fluoresces at 520nm (Ex. 500nm) when bound to double stranded DNA, and 650nm (Ex. 460nm) when bound to RNA or single-stranded DNA. As acridine orange is mutagenic, this product is supplied as a pre-made solution to prevent exposure during weighing.
Direction for Use
Adjust volume and staining time as necessary. Because some cells need to be fixed prior to staining, preliminary testing should be performed.
1. Remove medium from cells and wash twice with cold PBS(-).
2. Remove PBS(-) and add an appropriate volume of 1:50 Acridine Orange (Product No. A3396) in PBS(-). Allow to stain for 15 minutes.
3. Remove the staining solution and wash twice with PBS(-).
4. Observe cells with a fluorescence microscope while submerged in PBS(-).
Storage
Upon receipt the product should be stored at 4 °C, protected from light, and used within 6 months.
