Application
Tolcapone: A Selective and Reversible Inhibitor of Catechol-O-methyltransferase (COMT)
Tolcapone is a selective and reversible inhibitor of catechol-O-methyltransferase (COMT) which is one of several enzymes that degrade catecholamines (such as L-DOPA [D0600], dopamine [A0305], L-epinephrine [A0173], and norepinephrine [A0906]). COMT catalyzes the transfer of the methyl group of S-adenosyl-L-methionine to the phenolic group of substrates that contain a catechol structure including catecholamines. Tolcapone crosses the blood-brain barrier and can inhibit both peripheral and central COMT activity. Therefore, it has been suggested that tolcapone prolong the biological half-life of L-DOPA and the therapeutic effect of L-DOPA for Parkinson's disease patients.
