Description
- DiD is a lipophilic carbocyanine dye used to label cell membranes. It emits strong fluorescence when its long hydrocarbon chains are incorporated into a lipid bilayer. This product is a ready-to-use solution of DiD which can be used as a tracer in cell fusion, cell adhesion and chemotaxis.
Storage
- Store at room temperature and protect from light.
Directions for Use: Staining live cells

- Dilute DiD solution to 5 μM with culture medium.
- Remove medium from cells and replace with 5 μM DiD-containing medium.
- Incubate cells at 37 °C for 10 minutes.
- Remove medium and wash cells 3 times with PBS.
- Observe stained cell membranes via a fluorescence microscope.
Directions for Use: Staining fixed cells

- Remove culture medium and wash cells twice with PBS.
- Add 4% paraformaldehyde to cells and incubate at room temperature for 10 minutes.
- Dilute DiD solution to 5 μM with PBS.
- Remove 4% paraformaldehyde and add 5 μM DiD-containing PBS to cells.
- Incubate cells at 37 °C for 10 minutes.
- Remove medium and wash cells 3 times with PBS.
- Observe stained cell membranes via a fluorescence microscope.
