Nimustine (ACNU) hydrochloride (NSC 245382) is a water soluble DNA alkylating and crosslinking agent which induces DNA double-strand breaks (DSBs), but is not a simple alkylating agent. Thus, its exact mechanism of action has yet to be established. ACNU has been used for the treatment of gliomas. Recently, O6-methylguanine-DNA methyltransferase (MGMT) have received widespread attention. MGMT is a repair protein enzyme that specifically removes promutagenic alkyl groups from the O6 position of guanine in DNA. Repair of O6-alkylguanine adducts by tumour cells has been implicated in drug resistance since it reduces the cytotoxicity of alkylating agents including ACNU.
