Application
Oxyma: An Efficient Additive for Peptide Synthesis to Replace HOBt and HOAt
Experimental procedure4): a mixture of 1 (0.12 mmol), EDC·HCl (28 mg, 0.15 mmol, 1.2 eq.), Oxyma (20 mg, 0.15 mmol, 1.2 eq.), and DIEA (52 µL, 0.29 mmol, 2.4 eq.) in DMF (1.2 mL) is stirred for 15 min, and then 2 (0.12 mmol) is added. The mixture is stirred at room temperature for 20 hr and diluted with EtOAc. The organic phase is washed with 0.5 mol/L HCl, 5% LiCl, saturated aqueous NaHCO3, and brine, dried over Na2SO4 and concentrated in vacuo. The crude product is purified with silica column chromatography eluting with 3:2 hexanes:EtOAc to yield 3 (48 mg, 0.10 mmol, 82% yield) as a white solid.
