T2024 is used for the detection of free amino group on resin beads in Solid Supported Synthesis.
If resin beads react with T2024 to give a red color by the addition of a drop of T2024, the primary amines exist on resin beads.
Therefore T2024 does not suit for quantitative analysis, but only for qualitative analysis.
TNBS test is a safe and easy detection method of free amino group as compared with Kaiser test which requires potassium cyanide.
[Example of use]
Suspend some resin beads in DMF.
Add a drop of 1% Picrylsufonic Acid solution to resin beads suspension, followed by the addtion of a drop of 10% N,N-Diisopropylethylamine solution.
Leave for 5 minutes at room temperature.
Wash the resin beads with DMF and remove red solution.
If resin beads reddish in color, the primary amines exist on resin beads.
