Pregabalin: A Potent Ligand for the α2δ Subunit of Voltage-Gated Ca2+ Channels in the Central Nervous System
Pregabalin is one of the analogs of γ-aminobutyric acid (GABA) [A0282], but it works as a potent ligand for the α2δ subunit of voltage-gated Ca2+ channels in the central nervous system that exhibits potent anxiolytic, analgesic, and anticonvulsant properties in a range of animal models. Pregabalin binds potently and selectively to the α2δ subunit of "hyper-excited" voltage-gated calcium channels (VGCCs). The binding of pregabalin to VGCCs changes their conformation, reducing Ca2+ influx at nerve terminals. Consequently, pregabalin modulates the release of excitatory neurotransmitters in "hyper-excited" neurons, restoring them to normal physiological state. This mechanism of action is believed to confer on pregabalin its anxiolytic, analgesic, and anticonvulsant properties. (The product is for research purpose only.)
