Description
- This product is supplied as a pH-adjusted, 1 mg/mL stock solution of the linear form of the cationic polymer polyethylenimine (PEI) to be used for the transfection of nucleic acids into living cells.
Direction for Use

- Prepare two aliquots of OPTI-MEM®.
Each should be equivalent to 1/4 the regular culture maintenance volume of your cells. - Into one aliquot, mix an appropriate amount of Linear PEI HCl (Product No. L0466) (*).
- Into the other aliquot, mix an appropriate amount of DNA (*).
- Combine the two aliquots from step 2 and incubate at 37 °C for 20 minutes.
- Add an equivalent volume of regular culture medium to the solution from step 3.
- Aspirate medium from cells and replace with the medium from step 4.
- Allow transfection to proceed for between 6 and 24 hours.
- Replace medium and incubate until protein expression is detected.
- Cells can now be used in downstream applications.
- Prepare two aliquots of OPTI-MEM®.
- * Before running any transfection experiments, optimal transfection conditions (PEI concentration, N:P ratio) for your specific cell line must be determined. We recommend starting with final PEI concentrations ranging from 1 to 6 µg/mL, and N:P ratios of between 10 and 30.
