5'-Chloro-5'-deoxyadenosine: A Nucleoside Analog Used as a Substrate in Polyketide Biosynthesis
Incorporating a halogen atom into a scaffold of bioactive compounds often enhances biological activity, as in the case of the anticancer agent marizomib (salinosporamide A, a polyketide), a natural product with potent 20S proteasome inhibitory activity isolated from the marine microorganism Salinispora tropica. 5'-Chloro-5'-deoxyadenosine is a nucleoside analog used as a substrate in polyketide biosynthesis. S-adenosyl-L-methionine (SAM) is converted to 5'-chloro-5'-deoxyadenosine in a reaction catalyzed by SAM-dependent chlorinase. Through a 7-step route, 5'-chloro-5'-deoxyadenosine is converted to chloroethylmalonyl-CoA, which is involved in the biosynthesis of marizomib.
