Application
Stereoselective synthesis of cis-3,5-disubstituted morpholines

Typical procedure: A dried Schlenk tube is charged with Pd(OAc)2 (2.3 mg), tri(2-furyl)phosphine (9.3 mg), and NaOtBu (96.1 mg) under nitrogen. The tube is evacuated and backfilled with nitrogen, then 2-bromotoluene (171 mg) and a solution of (–)-(S)-N-[1-(allyloxy)-3-phenylpropane-2-yl]aniline (134 mg) in toluene (1.25 mL) are added to the tube. The mixture is heated to 105 °C for 18 h with stirring. The reaction mixture is cooled to rt, quenched with saturated aqueous NH4Cl (3 mL), and extracted with EtOAc (3 mL x 3). The combined organic layers are concentrated in vacuo and the crude product is purified by flash chromatography on silica gel (eluent: 5 % EtOAc/hexanes). This procedure gives (–)-(3S,5R)-3-benzyl-5-(2-methylbenzyl)-4-phenylmorpholine (121 mg, 68 %).
