CHIR 99021 is also used with XAV 939 as a key component for mouse epiblast stem cell and human embryonic stem cell self-renewal.¹⁾ Under cytokine-free conditions, the combination of CHIR 99021 and the mTOR inhibitor, rapamycin, maintains mouse and human hematopoietic stem cells.²⁾ In the presence of CHIR 99021, OCT4-transduced human blood CD34⁺ cells are converted into induced mesenchymal stem cells (iMSCs).³⁾

CHIR 99021: A Specific Glycogen Synthase Kinase 3 (GSK3) Inhibitor

CHIR 99021 is a specific glycogen synthase kinase 3 (GSK3) inhibitor. GSK3 is a serine/threonine kinase that mediates the addition of phosphate molecules onto serine and threonine amino acid residues, and that plays a key role in both the insulin and Wnt signaling pathways.^1,2) CHIR 99021 specifically inhibits the GSK3 pathway.^3,4)
