Procarbazine: An Antitumor Alkylating Agent
Procarbazine is an antitumor alkylating agent that was first synthesized in the late 1950s. Reportedly, procarbazine may inhibit transmethylation of methyl groups of methionine into t-RNA, and the non-functioning t-RNA could cause the cessation of DNA, RNA, and protein synthesis.
In addition, procarbazine can also directly damage DNA through an alkylation reaction. Alkylating agents including procarbazine are known to be associated with production of reactive oxygen species (ROS) which may attack protein sulfhydryl groups contained in protein which is tightly bound to DNA. Procarbazine is not cross-resistant with other alkylating agents. Procarbazine has been used in the treatment of many cancers, but its main use is in the treatment of Hodgkin's disease and brain tumors, since it can cross the blood-brain barrier.
