Description
This kit consists three reagents; 3, 3’-diaminobenzidine (DAB) solution, Hydrogen Peroxide solution and 1.5M Tris-HCl buffer and it is supplied to detect horseradish peroxidase (HRP) activity in Western blotting and so on. When DAB reacts with HRP, a brown precipitate appears. This kit does not include nickel component.
Direction for Use
1) Prepare a blotting membrane covered with HRP-conjugated antibody.
2) Prepare the DAB substrate solution according to the following volume;
- Add 5 drops of DAB solution (about 140 µL)
- Add 2 drops of Hydrogen Peroxide solution (about 80 µL)
- Add 2 drops of 1.5M Tris-HCl buffer (about 80 µL)
- Each reagent is added into 5 mL of deionized water
3) Soak the membrane in the DAB substrate solution and incubate until the membrane is colored.
4) Add deionized water and stop the coloring reaction.
Storage
The product should be stored at 4 °C and used within 6 months.
