Description
- This Endo-α is a glycoside hydrolase that recognizes the structure of the Galβ1-3GalNAc disaccharide α-linked with a hydroxyl group and it releases Galβ1-3GalNAc (galacto-N-biose) by hydrolysis. The Endo-α gene is derived from Bifidobacterium longum, and its recombinant protein is expressed in Escherichia coli.
Unit Definition
- One unit (U) of the Endo-α is defined as the amount required to produce 1 μmol of Galβ1-3GalNAc from Galβ1-3GalNAc α-pNP (Product No. G0375) per minute at 37°C under the condition of pH 5.0.
Assay Conditions
- Reaction conditions: 50 mM Sodium acetate, pH 5.0
The reaction mixture includes the enzyme and Galβ1-3GalNAc α-pNP in 20 μL. The mixture is incubated at 37°C for 10 minutes. After the enzyme reaction, quenching is performed by adding 30 μL of 0.25 M sodium carbonate. Quantification of released pNP is measured by a UV spectrometer.

- Store at -80°C in its lyophilized powder form. Use promptly after dissolving in buffer.
