Topotecan Hydrocholoride: A Water-Soluble Topoisomerase I Inhibitor
Topotecan hydrocholoride is a topoisomerase I inhibitor that is a semi-synthetic water-soluble derivative of camptothecin [C1495].1) During the course of its normal cell cycle, topoisomerase I relieves torsional strain in DNA by inducing reversible single strand breaks, and transiently forms a covalent bond with DNA. Topotecan prevents the re-ligation of these single-strand breaks by binding to and stabilizing the complex of topoisomerase I and DNA. This process leads to breaks in the DNA strand resulting in apoptosis and cell death.1-3) Recently, lipid nanoencapsulation of topotecan has attracted much attention in order to reduce the loss of efficacy of topotecan due to its hydrolysis in vivo4). (The product is for research purpose only.)
