Description

  • Exosomes are vesicles of 30-200 nm in diameter secreted by almost all cell types. They contain biologically important molecules (lipids, proteins, mRNAs, miRNAs) and are involved in many physiological and pathological processes. This product can be used to easily isolate exosomes from cell culture media. Isolated exosomes can be used in downstream analyses.

Direction for Use

    1. Collect cell culture media and centrifuge the media at 3,000 x g for 15 minutes at 4°C.
    2. Transfer the supernatant to a new tube.
    3. Add Exosome Isolation Reagent [for Cell Culture Media] to the supernatant and mix via pipetting until homogeneous. Use 500 μL of reagent per 1 mL supernatant.
    4. Incubate at 4°C overnight.
    5. Centrifuge at 10,000 x g for 1 hour at 4°C.
    6. Remove the supernatant carefully (pellet is often invisible).
    7. Resuspend pellet in 1x PBS or another specific buffer. Use 25-50 μL of buffer per 1 mL of starting media.
    8. Exosomes are now ready to be used in further experiments.

Storage

  • Store at 4 °C and use within 6 months.
