Synthesis and metabolic evaluation of azaspirocyclic fluoroquinolones
Typical procedure: KOtBu (191 mg) is dissolve in DMSO (20 mL), azetidinium oxalate (164 mg) is added and the mixture is stirred at rt for 10 min. Then, 7-chloro-1-cyclopropyl-6-fluoro-1,4-dihydro-4-oxoquinoline-3-carboxylic acid (200 mg) is added, and the yellow mixture is heated to 130 °C and stirred for 5.5 h. Then it is cooled to rt and poured into H2O (75 mL). The precipitate is filtered and dried overnight upon standing. The crude product is purified by suspending in hot CHCl3, and cooling the mixture to rt, when it is filtered. The residue is dried in vacuo to give the pure desired product (166 mg, 68 %).
