Clioquinol: A Metal-Chelating Antimicrobial Agent with Neuroprotective Properties
Clioquinol was first developed in 1930s as a topical antiseptic and an oral an oral intestinal amebicide. In the early 1970s, clioquinol was withdrawn from the pharmaceutical market as an oral agent due to its association with subacute myelooptic neuropathy (SMON). Recently, clioquinol has been reported as a metal-chelating antimicrobial agent with neuroprotective properties. Metal chelation is a potential therapeutic strategy for Alzheimer's disease (AD) because zinc and copper are involved in the deposition and stabilization of β-amyloid plaques, and chelating agents can dissolve amyloid deposits in vitro and in vivo.
