Amodiaquine Dihydrochloride: A 4-Aminoquinoline Antimalarial Agent

Amodiaquine dihydrochloride is a synthetic 4-aminoquinoline antimalarial agent. Amodiaquine is mainly metabolized hepatically towards its major active metabolite N-desethylamodiaquine, by the cytochrome P450 2C8 (CYP2C8). The mechanism of action of amodiaquine against plasmodium is not yet completely understood, but the most plausible explanation is that the active metabolite penetrate the infected red blood cells and prevent the parasite from polymerizing heme into a malarial metabolite called hemozoin, leading to parasite death. Recently, the antimalarial combination of amodiaquine plus artesunate [A2191] has been extensively studied and good efficacy and tolerability has been reported.
