Activation of AMP-activated Protein Kinase (AMPK) by Metformin

Metformin hydrochloride, an oral biguanide antihyperglycemic agent, is not chemically or pharmacologically related to any other classes of oral antihyperglycemic agents. Metformin lowers the blood glucose concentration without causing hypoglycemia, but the precise mechanism of action of metformin is not fully understood. However, it has been reported that metformin activates adenosine 5’-monophosphate-activated protein kinase (AMPK, a major cellular regulator of lipid and glucose metabolism) in hepatocytes. As a result, hepatic glucose production is decreased and skeletal myocyte glucose uptake is increased. Recently, some combination agents with other anti-diabetes agents (eg. sulfonylureas and thiazolidinediones) have been developed to treat type-2 diabetes. (The product is for research purpose only.)
