A Fluorescent Probe for Thiol-Specific Labeling
Bromobimane (mBBr) is a fluorescent probe linking specifically to thiol groups, which has a maximum absorption at about 390 nm with the labeled products producing fluorescence at about 480 nm (maximum wavelength).1) For example, mBBr has been applicable to label biological materials such as cysteine, glutathione, coenzyme A, and proteins including thiol groups.2-4) In protein research, it has been revealed that the status of disulfide bonds, formed by a combination of cysteine residues, regulates the functional activity of proteins (redox regulation). So proteomic analysis with the use of mBBr has been performed.5) Furthermore, mBBr-labeling has been applied to the analysis of proteins in food.6)
