Description
- This product is supplied as a ready-to-use solution to lyse cultured mammalian cells. The extraction from cultured cells by RIPA buffer (Product No. R0246) can be transferred directly to further analysis. This product does not include protease inhibitors, thus please add protease inhibitor cocktail to the product as necessary.
Direction for Use: Lysis of adherent cells

- Remove medium from cultured cells and wash cells twice with cold PBS.
- Remove PBS and add cold RIPA buffer (Product No. R0246) to the cells. 1 mL of RIPA buffer is added to 0.5 - 5.0 x 107 cells.
- Scrape the adherent cells with a cell scraper and transfer the lysate and pellet to a tube.
- Incubate the cells for 15 minutes on ice. (To increase the yields, prolong the incubation.)
- Centrifuge the tube at 10,000 x g for 10 minutes at 4 °C.
- Transfer the supernatant to a new tube for further analysis.

- Transfer the cultured cells to a tube, remove medium from cultured cells and wash cells twice with cold PBS.
- Remove PBS and add cold RIPA buffer (Product No. R0246) to the cells. 1 mL of RIPA buffer is added to 0.5 - 5.0 x 107 cells.
- Incubate the cells for 15 minutes on ice. (To increase the yields, prolong the incubation.)

- Transfer the supernatant to a new tube for further analysis.
Storage
- The product should be stored at 4 °C and used within 6 months.
