P3S is an RNA structural mapping reagent that sulfonylates the 2'-OH groups of ribonucleotides in unpaired regions of RNA.1) P3S has advantages such as good solubility in aqueous solvents and a long half-life in water. In addition, P3S shows a different modification pattern from SHAPE (Selective 2'-Hydroxyl Acylation and Primer Extension) reagents,2) which acylate 2'-OH groups like P3S and have been employed for the analysis of RNA structural characteristics. Thus, it is expected that a new structural analysis using P3S will lead to the development of applications for RNA.
