The phosphoramidite method is a general-purpose oligonucleotide synthesis method used in nucleic acid synthesis research. Condensation activators are reagents used in the phosphoramidite method for the coupling reaction between the 5' position hydroxyl group at the end of the oligonucleotide chain and the nucleoside phosphoramidite. Various types of tetrazole, imidazole, and imidazolium salts are available so that the acidity and nucleophilicity can be changed to match the coupling substrate, and condensation activators that satisfy both phosphoramidite activation and detritylation inhibition can be selected.
