Pioglitazone Hydrochloride: An Agonist at the Peroxisome Proliferator-Activated Receptor of the Gamma Subtype (PPARγ)

Pioglitazone hydrochloride, a thiazolidinedione, is an agonist at the peroxisome proliferator-activated receptor of the gamma subtype (PPARγ), a transcription-regulating receptor. PPAR receptors are found in peripheral tissues important for insulin action. Activation of PPARγ nuclear receptors modulates the transcription of a number of insulin responsive genes involved in the control of glucose and lipid metabolism. Therefore, pioglitazone decreases insulin resistance in the periphery tissues and in the liver resulting in increased insulin-dependent glucose disposal and decreased hepatic glucose output. And recently, it has been reported that pioglitazone slows down the progression of Alzheimer's disease. For your reference, [A2567] is a key synthetic intermediate of pioglitazone.
