1-Adamantanamine Hydrochloride: A Dopamine Receptor Agonist and an Anti-Influenza A Virus Agent
1-Adamantanamine hydrochloride and its free base [A2318] are dopamine receptor agonists and anti-influenza A virus agents, similarly rimantadine hydrochloride [R0070]. 1-Amantadine was originally developed as an antiviral agent. One day, a single patient noticed relief in her Parkinson’s disease symptoms after taking amantadine for a flu infection, and this observation led to a new drug indication.1)
These mechanisms of action are not fully understood. However, there is evidence to suggest that 1-adamantanamine act as a dopamine receptor agonist by enhancing the synthesis and release of dopamine from nerve cells and delaying the reuptake into synaptic vesicles.2,3)
In addition, 1-adamantanamine inhibits the replication of influenza A viruses by preventing the release of infectious viral RNA into the host cell nucleus by interfering with the function of the transmembrane of the viral M2 protein.4,5) (The product is for research purpose only.)
