Plerixafor: A CXCR4 Chemokine Receptor Inhibitor and a Stem Cell Mobilization Agent
Plerixafor (AMD1300) was originally discovered as an anti-HIV agent effective in inhibiting the replication of HIV.1) However, at present, plerixafor has been used as a CXCR4 chemokine receptor inhibitor and a hematopoietic stem cell mobilization agent.2,3) Hematopoietic stem cells express CXCR4, a seven-transmembrane G-protein-coupled chemokine receptor. Plerixafor works as a CXCR4 chemokine receptor antagonist and blocks binding of its cognate ligand, stromal cell-derived factor-1α (SDF-1α). CXCR4 with its ligand SDF-1α has been reported to play an important role in engraftment of hematopoietic stem cells (HSCs) in the bone marrow. Plerixafor results in the rapid and reversible mobilization of hematopoietic stem cells into the peripheral circulation. (The product is for research purpose only.)
