Description

  • DiI is a lipophilic carbocyanine dye used to label cell membranes. It emits strong fluorescence when its long hydrocarbon chains are incorporated into a lipid bilayer. This product is a ready-to-use solution of DiI which can be used as a tracer in cell fusion, cell adhesion and chemotaxis.

Storage

  • Store at room temperature (below 15 °C is recommended) and protect from light.

Directions for Use: Staining live cells

    1. Dilute DiI solution to 5 μM with culture medium.
    2. Remove medium from cells and replace with 5 μM DiI-containing medium.
    3. Incubate cells at 37 °C for 10 minutes.
    4. Remove medium and wash cells 3 times with PBS.
    5. Observe stained cell membranes via a fluorescence microscope.

Directions for Use: Staining fixed cells

    1. Remove culture medium and wash cells twice with PBS.
    2. Add 4% paraformaldehyde to cells and incubate at room temperature for 10 minutes.
    3. Dilute DiI solution to 5 μM with PBS.
    4. Remove 4% paraformaldehyde and add 5 μM DiI-containing PBS to cells.
    5. Incubate cells at 37 °C for 10 minutes.
    6. Remove medium and wash cells 3 times with PBS.
    7. Observe stained cell membranes via a fluorescence microscope.
