Application
p-Toluenesulfonylmethyl Isocyanide Derivatives Effective in the Construction of Heterocyclic Compounds
Experimental procedure: To 4-pentenal (3, 252 mg, 3.0 mmol) in DMF (3 mL) is added allylamine (2, 171 mg, 3.0 mmol), and the reaction mixture is stirred at room temperature for 2.5 h. This is followed by the addition of α-(p-toluenesulfonyl)benzyl isocyanide (1, 543 mg, 2.0 mmol) and K2CO3 (276 mg, 2.0 mmol), and the reaction mixture is allowed to stir for an additional 17 h at room temperature. The reaction is quenched by the addition of water. The aqueous layer is extracted with EtOAc, dried (anhydrous MgSO4), concentrated, and purified by flash column chromatography (CH2Cl2:CH3OH:NH3 = 97:2.5:0.5) to afford 433 mg (91%) of 4 as an oil.
