Rucaparib: An Inhibitor of Poly (ADP-Ribose) Polymerase (PARP) Enzymes

Genomic instability is a hallmark of cancer and is often the result of altered DNA repair capabilities in tumor cells. Poly (ADP-ribose) polymerases (PARPs) are enzymes responsible for DNA damage detection and repair factor recruitment. PARP inhibitors are able to interact with the binding site for the PARP cofactor NAD+ and trap PARP on DNA, and induce cell death through replication stress-induced mitotic catastrophe. Rucaparib (AG014699; PF-01367338) has been reported as an inhibitor of PARP enzymes, including PARP-1, PARP-2 and PARP-3, which play a role in DNA repair. In vitro studies have shown that rucaparib-induced cytotoxicity may involve inhibition of PARP enzyme activity and increased formation of PARP-DNA complexes resulting in DNA damage, apoptosis, and cancer cell death.
