Application
A Guanidinylating Reagent
Experimental procedure: to a suspension of N-(tert-butoxycarbonyl)-1H-pyrazole-1-carboxamidine (3.15 g, 15 mmol, 1 eq.) and aniline hydrochloride (5.84 g, 45 mmol, 3 eq.) in THF (14 mL) is added DIEA (8.55 mL, 49 mmol, 3.3 eq.). The reaction mixture is stirred under reflux under argon for 3 days. After removal of the solvent under reduced pressure, the product is purified by flash chromatography on silica gel (DCM saturated with ammonia/MeOH: 99/1 then 98/2) to yield the corresponding product (2.3 g, 67% yield) as a white solid.
