Toepassing
Lonidamine: An Anticancer Agent Acting on Mitochondria
Lonidamine, a derivate of indazole-3-carboxylic acid, was initially developed as an anti-spermatogenic agent. It was later reported to have anticancer activity with a peculiar mechanism of action. Lonidamine has no function on cellular nucleic acids or protein synthesis, whereas acts on mitochondria to induce apoptosis. Thus, lonidamine exerts a powerful inhibitory effect on oxygen consumption, aerobic glycolysis and lactate transport and accumulation of neoplastic cells. Although the clinical development of lonidamine have been discontinued, it has recently attracted attention as a combination agent with other antitumor agents.
